The newborn gut undergoes rapid colonization by commensal microorganisms and possible exposure to pathogens. The contribution of colostrum intake to host protection is well known; however, limited research exists on the intestinal innate immunity corresponding to colostrum intake during the passive immune transfer period in newborn ruminants. The aim of this study was to investigate the changes in bacterial community and expression of genes encoding toll-like receptors (TLR), mucins (MUC), antimicrobial peptides, and tight junctions in the jejunum of lambs that were fed colostrum during the first 24 h of life. Twenty-seven newborn lambs were used in this study, of which 18 lambs were bottle-fed pooled bovine colostrum within the first 2 h after birth to obtain an intake of approximately 8% of body weight. Lambs were slaughtered at 12 (n = 9) and 24 h (n = 9) after birth. The remaining 9 lambs without any feeding were slaughtered at 30 min after birth (0 h). Tissue and ligated segment samples from the jejunum were collected immediately after the lambs were slaughtered. The bacterial profile in the ligated jejunum segment was assessed using amplicon sequencing. The gene expression in the jejunum tissue was determined using quantitative real-time PCR. The relative abundances of Escherichia-Shigella, Lactobacillus, Lactococcus, and Streptococcus increased, whereas those of Sphingomonas, Phyllobacterium, Bradyrhizobium, and Rudaea decreased during the first 24 h of life. Expression of TLR2 and β-defensin 109-like was upregulated at 12 h after birth, but a recovery was detected at 24 h; TLR3, TLR5, LYZ, MUC1, MUC13, MUC20, and CLDN7 showed a higher expression level in samples taken at 24 h than in those taken at 0 h. In addition, expression level of CLDN1, CLDN4, and the junctional adhesion molecule-1 tended to be higher at 24 h than at 0 h after birth. Correlation analysis indicated that TLR2 expression was negatively correlated with the relative abundance of Lactobacillus and Bradyrhizobium, whereas TLR5 expression was positively correlated with the relative abundance of Escherichia-Shigella and Pelagibacterium. These results suggest that TLR, MUC, antimicrobial peptides, and CLDN act together and play an important role in intestinal defense during the passive immune transfer period. They are potentially associated with microbial colonization. The findings from this study provide novel information to elucidate the role of colostrum components in regulating the development of the intestinal mucosal immune barrier in newborn lambs during the passive immune transfer period.  相似文献

Biochar has been used as an environment-friendly enhancer to improve the hydraulic properties(e.g.suction and water retention) of soil.However,variations in densities alter the properties of the soil-biochar mix.Such density variations are observed in agriculture(loosely compacted) and engineering(densely compacted) applications.The influence of biochar amendment on gas permeability of soil has been barely investigated,especially for soil with diffe rent densities.The maj or obj ective of this study is to investigate the water retention capacity,and gas permeability of biochar-amended soil(BAS) with different biochar contents under varying degree of compaction(DOC) conditions.In-house produced novel biochar was mixed with the soil at different amendment rates(i.e.biochar contents of 0%,5% and 10%).All BAS samples were compacted at three DOCs(65%,80% and 95%) in polyvinyl chloride(PVC)tubes.Each soil column was subjected to drying-wetting cycles,during which soil suction,water content,and gas permeability were measured.A simplified theoretical framework for estimating the void ratio of BAS was proposed.The experimental results reveal that the addition of biochar significantly decreased gas permeability k_g as compared with that of bare soil(BS).However,the addition of 5%biochar is found to be optimum in decreasing kg with an increase of DOC(i.e.k_(g,65%) k_(g,80%) k_(g,95%)) at a relatively low suction range(200 kPa) because both biochar and compaction treatment reduce the connected pores.  相似文献

The cobalt-free cathode materials with high activity is critical to the commercial application of medium temperature solid oxide fuel cells (IT-SOFC). Herein, a series of cobalt-free Nb-doped Pr2Ni1-xNbxO4 (x = 0, 0.05, 0.10, and 0.15) perovskite oxides were successfully prepared, and the effects of Nb-doping on the structure, thermal stability and electrochemical properties of the cathode material are studied in detail. The Pr2Ni1-xNbxO4 exhibits a K2NiF4 type structure with Fmmm space group. The Nb5+ cations that doped into Pr2NiO4 replaces the Ni site, which increases the surface oxygen content, then effectively eliminates the phase transition of Pr2NiO4 and significantly improves the ORR catalytic activity. The Pr2Ni0.9Nb0.1O4 was found to occupy the lowest polarization resistance of 0.057 Ω cm2, and the peak power density of single cells supported by the electrolyte is 0.576 W cm?2 at 700 °C, which has good long-term stability.  相似文献

In this article, the nonlinear dynamic characteristics and bifurcation of a Ti–Zr–Ni quasicrystal impacted by hydrogen atoms are studied. New nonlinear damping terms are proposed to express the delay characteristics of Ti–Zr–Ni quasicrystal, and the accurate natural frequency is obtained by the harmonic balance method. A new method based on the developed largest Lyapunov exponent is proposed to analyze the local stability of any point in the system, and the system's global stability is determined. Finally, a new way to realize the switch between hydrogen storage and release based on stochastic Hopf bifurcation is proposed. The results of theoretical analysis and numerical simulation show that the system's motion can be switched between a periodic orbit and a balanced point near the bifurcation boundary with little energy consumption, which is helpful for hydrogen storage and release.  相似文献

We designed and fabricated non-precious and highly efficient electrocatalysts of nickelmolybdenum/copper-nanosheets/nickel-foam composites (NiMo/Cu-NS/NF) by step electrodepositions, combining with chemical oxidation method. The catalysts were charaterized by means of SEM, XRD and XPS spectra. Their electrocatalytic activities were assessed by hydrogen evolution reactions (HER) over a wide pH range, where acidic, neutral and alkaline electrolytes were used, respectively. Benefiting from the unique midlayer Cu nanosheets (NS) architecture and optimum Mo–Ni composition at the surface layer which led to high electronic conductivity and large electrochemically active surface area (ECSA), the NiMo/Cu-NS/NF-2 catalyst displayed superior electrocatalytic activities with low overpotentials of η10 = 43, 86 and 89 mV in 0.5 M H2SO4, 1.0 M PBS and 1.0 M KOH electrolyte, respectively. Especially in the acidic condition, it exhibited the best electrocatalytic activity with smaller Tafel slope of 54 mV dec?1 and higher exchange current density of 1.93 mA cm?2.  相似文献

To accelerate the commercialization of fuel cells, many efforts have been made to develope highly active and durable Pt-based catalyst for oxygen reduction reaction (ORR). Herein, PtCu porous nanowires (PNWs) with controllable composition are synthesized through an ultrasound-assisted galvanic replacement reaction. The porous structure, surface strain, and electronic property of PtCu PNWs are optimized by tuning composition, which can improve activity for ORR. Electrochemical tests reveal that the mass activity of Pt0.5Cu0.5 PNWs (Pt/Cu atomic ratio of 1:1) reaches 0.80 A mgPt?1, which is about 5 times higher than that of the commercial Pt/C catalyst. Notably, the improved activity of the porous nanowire catalyst is also confirmed in the single-cell test. In addition, the large contact area with the carrier and internal interconnection structure of Pt0.5Cu0.5 PNWs enables them to exhibit much better durability than the commercial Pt/C catalyst and Pt0.5Cu0.5 nanotubes in accelerated durability test.  相似文献

Laminarin and its derived oligosaccharides have diverse bioactivities. The β-1,3-glucanase in marine bacteria can be employed as a tool to digest laminarin in the cell wall of brown algae. Here, we cloned, expressed and characterised a β-1,3-glucanase (laminarinase), MaLamNA, from the previously characterised marine bacterium Microbulbifer sp. ALW1, phylogenetically distinct from the glycoside hydrolase families of characterised laminarinases. The recombinant laminarinase was heterologously expressed and purified from Pichia pastoris GS115 cells with a molecular mass of 57.3 kDa. MaLamNA exerted its hydrolytic activity specifically against laminarin, with the highest activity at 45 °C and pH 4.5–5.5, respectively, and demonstrated high stability against extreme acidic and alkaline pH exposure. The addition of reducing agent dithiothreitol could significantly enhance the activity of MaLamNA. The hydrolytic products of laminarin by MaLamNA exhibited more effective antioxidant activities than the undigested laminarin. These characteristics of MaLamNA provide clues to its industrial application for laminarin bioresource development.  相似文献

It is essential to develop an accurate model of proton exchange membrane fuel cell (PEMFC) for a reliable operation and analysis, in which unknown parameters usually need to be determined. The inherent nonlinear, strong coupling, and diversification of PEMFC model seriously hinder traditional methods to identify the parameters. For the sake of overcoming these thorny obstacles, Levenberg-Marquardt backpropagation (LMBP) algorithm based on artificial neural networks (ANNs) is proposed for PEMFC parameter identification. Furthermore, the performance of LMBP is thoroughly evaluated and compared with four typical meta-heuristic algorithms under three cases. Simulation results indicate that LMBP performs a higher accuracy and faster speed for parameter identification. In particular, accuracy and convergence speed can achieve as much as 99.8% and 95.9% growth via LMBP, respectively.  相似文献

This article develops an approximation-based fuzzy control scheme for nonstrict feedback stochastic nonlinear systems (NFSNS) with time-varying state constraints. The difficulty in constructing controller is how to conquer the algebraic loop problem caused by nonstrict feedback structure, as well as prevent the state constraints from violating. To dispose the time-varying state constraints, time-varying barrier Lyapunov function is incorporated into the backstepping design framework. The lumped uncertainties of NFSNS are approximated by the fuzzy logic systems. By virtue of fuzzy basis function, the algebraic loop problem is effectively handled. Theoretical analysis shows that the predefined state constraints are not violated and all signals of the closed-loop systems are bounded. Finally, simulation results substantiate the validity of the devised method.  相似文献
